XtenCHO ™ Cell Bank Pack
The XtenCHO™ Cell line has been an internal tool for years at ProteoGenix! These extremely powerful cells are now available for you! In the last years, biotherapeutics represent more than half of the total approved drugs. Most of them include monoclonal antibodies. Biotherapeutics bear post-translational modifications, are often large and complex. A stable cell line like XtenCHO™ which is able to produce these proteins is a valuable tool for therapeutic development. ProteoGenix uses this cell line routinely to produce its recombinant protein for its customers. Given the willingness of our customers to use it we now offer it in our catalog.
The XtenCHO™ Cells are derived from Chinese Hamster Ovarian CHO-K1 cell line. They have been optimized to sustain extended transient gene expression. The genetic modifications enhance plasmid-driven expression and increase plasmid maintenance. Indeed, the XtenCHO™ Cells can maintain the transfected plasmid for 14 days without feeding making them produce on a longer period. The XtenCHO™ Cells are well adapted to high-density suspension culture. The XtenCHO™ Expression Medium has been specifically developed to be supplied to these cells for high viability and high growth cultures. The XtenCHO™ Cells have a doubling time of approximately 23 hours. These cells are also very robust and easy to cultivate, their recovery time after thawing is about 2 days. The XtenCHO™ Cells provide the highest protein expression level on the market! In combination with the XtenCHO™ Transfection Kit, the record yield of 904 mg/mL has been achieved with a recombinant antibody. Moreover, these cells can produce a wide range of proteins and antibodies including pentameric IgMs. Being derived from CHO cells they perform human-like glycosylations and are resistant to viral infections. The XtenCHO™ Cells are key components of the XtenCHO™ Starter Kit. Cells are also provided in 6 vials of 1mL format in the XtenCHO™ Cell Bank. Therapeutic antibodies from several IgG subclasses and isotypes (IgG1-kappa and lambda, IgG2-lambda, IgG4-kappa) were produced with XtenCHO ™ cells and in parallel with CHO-S ™ and ExpiCHO ™ expression systems. XtenCHO ™ cells were transfected with ProteoGenix' proprietary TGE system (transfection protocol, transfection reagent and culture medium). CHO-S ™and ExpiCHO ™ cells were transfected according to manufacturer's instructions for each expression system (protocol, reagents and culture medium). Determination of production yields was done with an Octet Red96 system (Protein G biosensor).
